The manuscript "Infection Counter: Automated Quantification of in Vitro Virus Replication by Fluorescence Microscopy" presents the development and validation of Infection Counter, a novel, automated method for estimating the proportion of virally infected cells using fluorescence microscopy. This open-source ImageJ plugin is adaptable for various viruses and offers advantages over existing techniques, including simplicity, accessibility, and increased throughput. The authors demonstrate Infection Counter's equivalence to the focus-forming assay for hepatitis C virus (HCV) and apply it to evaluate the anti-HCV activity of a cyclosporine analogue, SMBz-CsA. The study suggests that SMBz-CsA may inhibit HCVcc replication through CypA-NS5A interactions and possibly other mechanisms. Overall, Infection Counter provides a reliable, medium/high-throughput assay for basic and translational research.
