Unveiling live-cell viral replication at the nanoscale

Proposes to use super-resolution microscopy and machine learning to track viral replication dynamics in living cells over long time periods, with the spatial and temporal resolution to observe key events like viral entry, uncoating, translation, assembly, and budding. The aim is to develop an adaptive, multimodal microscope that can image at different resolutions and switch modalities depending on the infection stage, while minimizing phototoxicity. This will be applied to study open questions in HIV-1 infection like how entry pathways affect downstream replication and which virions produce productive infections. The technology developed will be open-source to make it accessible for studying other viruses like influenza and SARS-CoV-2.
